Tenaya is dedicated to addressing heart failure through multipronged efforts that target the fundamental cellular pathologies present in diseased cardiac muscle and that leverage cutting-edge research in cardiac development and regeneration.

Tenaya has a lead gene therapy program focused on in vivo reprogramming of cardiac fibroblasts to cardiomyocytes to regenerate heart tissue, and additional programs focused on cardiomyopathies.

Role

We are seeking a motivated Research Associate for our Technical Operations, Process Development Group. In this role, you will perform key production and characterization activities to support Upstream Process Development; troubleshoot and optimize production unit operation and assays as needed; assist in the development of new characterization methods to determine precursor and in-process material purity and identity; and perform other tasks such as lab equipment maintenance and supply ordering.

Key Responsibilities
  • Routine support of production activities for rAAV manufacturing (benchtop reactor to pilot scale)
  • Routine application of molecular techniques including PCR, electrophoresis, restriction digestion, identity mapping, DNA isolation, and tittering
  • Cell Culture of multiple cell lines
  • Perform transformation, clone selection, and production by fermentation
  • Troubleshooting, optimization, and tracking of productions for both DNA and rAAV product
  • Assist in the development of new characterization and production methods in collaboration with supervisor
  • Documentation including, writing/reviewing of technical procedures and presentation of results in team meetings.
  • Maintain necessary laboratory notebook, supplies, equipment and space, prepare media and buffers.
  • Contribute to routine laboratory and instrument maintenance
  • Open minded attitude that start-up team environments require an assortment of skill and a high level of commitment

Requirements

  • 6+ months experience in related field or university research program
  • Knowledge of general (bio) chemistry, cellular and molecular biology laboratory techniques
  • Excellent aseptic techniques and experience with cell culture
  • Good oral and written communication
  • Ability to function well as part of a small dedicated team
  • Attention to detail
  • Demonstrated ability to identify process or protocol bottlenecks and variability and troubleshoot in collaboration with supervisor
  • The ability to work in a fast-paced team environment and to meet challenging deadlines under pressure

Preferred Skills & Competencies
  • Familiarity with the production or testing of viral vectors for gene therapy applications.
  • Familiarity with one or more of the following techniques: qPCR and/or ddPCR, ELISAs, PCR, DNA isolation, Electrophoresis (Agarose or PAGE), and Sequencing
  • Experience with cloning techniques and plasmid assembly
  • Experience with suspension cell culture in stirred tank bioreactor
  • Experience in writing technical procedures
